Investigation of Changes in Apoptosis and Adipokine Biomarkers in Rats With Experimental Hypothyroidism

Objective: In this study, it was aimed to investigate the changes in apoptosis and adipokine biomarkers in rats with propylthiouracil (PTU)-induced hypothyroidism.

Materials and Methods: This study was carried out on 14 Sprague–Dawley male rats. While 10 mg/kg PTU was given intraperitoneally to the hypothyroid group for 15 days, physiological saline was also applied to the control group. At the end of the study, heart and serum samples were taken from the sacrificed animals. Thyroid-stimulating hormone, fT4, irisin, adiponectin, BCL2, and TP53 levels were determined in these samples by enzyme-linked immunosorbent assay.

Results: Although body weights increased in both groups on day 15 of the study, this increase was significant only in the hypothyroid group (P = .002). The body weights of the hypothyroid group on the 15th day were higher than the control group (P = .048). Serum thyroid-stimulating hormone levels of the hypothyroid group (P = .000) were higher than the control group, and fT4 levels (P = .000) were lower. It was observed that irisin levels increased in hypothyroidism (P = .000), but ADP levels did not change. In addition, it was determined that BCL2 level, one of the important antiapoptotic markers, decreased in the hypothyroid group (P = .039), whereas the proapoptotic TP53 level increased, but this increase was not statistically significant.

Conclusion: It suggests that the increase in irisin levels determined in the heart tissue may be due to cardiac hypertrophy induced by hypothyroidism and that abnormal changes in the levels of thyroid hormones may induce apoptosis via the intrinsic pathway by reducing the level of antiapoptotic signaling proteins such as BCL2 by disrupting cell metabolism and cycle.

Deneysel Hipotiroidizm Oluşturulan Sıçanlarda Apoptoz ve Adipokin Biyomarkerlarındaki Değişikliklerin İncelenmesi

Amaç: Bu çalışmada propylthiouracil (PTU) ile indüklenen hipotiroidizmli sıçanlarda apoptoz ve adipokin biyomarkerlarındaki değişikliklerin incelenmesi amaçlanmıştır.

Gereç ve Yöntemler: Bu çalışma, 14 adet Sprague-Dawley erkek sıçan üzerinde gerçekleştirildi. Hipotiroidi grubuna 15 gün boyunca 10 mg/kg PTU intraperitoneal (i.p.) olarak verilirken; Kontrol grubuna da serum fizyolojik uygulandı. Çalışma sonunda sakrifiye edilen hayvanlardan kalp ve serum örnekleri alındı. Bu örneklerde Tiroit stimülan hormon (TSH), Serbest tiroksin (fT4), İrisin, Adiponektin (ADP), B-hücreli lenfoma-2 (BCL2) ve Tümör proteini P53 (TP53) düzeyleri eliza yöntemiyle belirlendi.

Bulgular: Çalışmanın 15. gününde vücut ağırlıkları her iki grupta da yükselse de bu yükseliş yalnızca Hipotiroidi grubunda önemliydi (P = ,002). Hipotiroidi grubunun 15. gündeki vücut ağırlıkları kontrole kıyasla yüksek bulundu (P = .048). Hipotiroit grubunun serum TSH düzeyleri (P = .000) kontrole göre yüksek, fT4 düzeyleri (P = .000) ise düşüktü. İrisin düzeylerinin Hipotiroidide arttığı (P = .000) fakat ADP düzeylerinin ise değişmediği gözlendi. Ayrıca önemli antiapoptotik markerlardan biri olan BCL2 seviyesinin Hipotiroidili grupta azaldığı (P = .039), buna karşın proapoptotik TP53 seviyesinin ise arttığı fakat bu artışın istatistiksel
olarak önemli olmadığı belirlendi.

Sonuç: Kalp dokusunda belirlenen irisin seviyelerindeki artışın hipotiroidizmin indüklediği kalp hipertrofisinden kaynaklanabileceğini ayrıca tiroit hormonlarının seviyelerindeki anormal değişimlerin hücre metabolizmasını ve siklusunu bozarak BCL2 gibi antiapoptotik sinyal proteinlerinin seviyesini azaltmak suretiyle intrinsik yolak üzerinden apoptozu indükleyebileceğini düşündürmektedir.
